There is a lot of information to consider when first getting started and one solution does not fit all, so we started wiki of known hardware configurations, and are encouraging the development community to add in their solutions/usage:

Searching prior mailing list Q/A or asking a new question about how others are doing this is also a great option. Please see the galaxy-dev@bx.psu.edu mailing list (and consider subscribing). Posting this question there, with a bit more detail about how many users, the types of jobs/volume, and special considerations such as budget, security, etc. , will likely get you some feedback from others already running instances under similar conditions.
